The Mark Twain House was designed by Edward Tuckerman Potter and built in 1874 in the High Victorian style. It served as the Twain family home until 1891, and it is now a National Historic Landmark. It contains an important collection of fine and decorative arts, including the only remaining domestic interiors by Louis Comfort Tiffany.

The Gallery sponsors three exhibitions per year of the work of professional artists from outside the Trinity community. In addition, there are two student shows and one faculty show each year. The permanent collection is spread around the campus.
